Vernonia amygdalina possess specialized metabolites which have been shown to minimize the risk of human diseases. This study was aimed to evaluate the effect of V. amygdalina extracts on Superoxide dismutase (SOD) and Glutathione s-transferase (GST) on alloxan induced diabetes in Wistar rats. Extraction of the V. amygdalina leaves was done manually and extracts were fractionated using Asymmetric Flow Field Flow Fractionation method. The fractions obtained from the crude V. amygdalina were tested to identify phytochemicals such as alkaloids, glycosides and saponins. Meanwhile, the experimental animals were divided into 2 phases. Phase 1 (non-induced) were grouped into 6 groups (control, crude, metformin, saponin, glycoside and alkaloid) of six animals. Phase 2 were grouped into 5 groups (crude, metformin, saponin, glycoside and alkaloid) of 6 animals which were induced intraperitoneally with Alloxan using injection (Alloxan-Induced). The results for the fasting blood glucose level of treated alloxan-induced diabetic showed a significant increase compared to those in control group (61.33±2.0) of the untreated. In addition, the histopathological analysis of the pancreas showed significant resurgence of destroyed pancreatic islet cells. The findings of the current research showed that Vernonia amygdalina ameliorated the enzymatic activities on alloxan induced diabetes on male Wistar rats.
