Abstract

Extended Data accompanying the F1000Research Study Pre-Protocol: "Systemic Calcification Phenotype: A Compositional Framework for Cross-Compartment Ectopic Mineralisation — A Program Architecture for Staged Empirical Stress-Testing." This document contains three extended data components: 1. H-PLS-FAM Specification — Full formal specification of the multi-block hierarchical Partial Least Squares Feature Aggregation Model, including measurement models, scaling constraints, identifiability conditions, sign orientation rules, and concordance assessment procedures. 2. Phase A1 Evidence Grading Protocol — Complete protocol for the systematic scoping review, including search strategy syntax for four databases (PubMed, Scopus, Web of Science, Embase), inclusion/exclusion criteria, data extraction form, detailed decision rule hierarchy with exemplar applications, and directionality confidence scoring criteria. 3. Illustrative Provisional Biomarker Categorisation — Provisional promoter–inhibitor assignments based on current mechanistic rationale, with full directionality justification for each biomarker. This catalogue carries an explicit disclaimer that it does not constitute the instantiated CSI₀ domain and is provided for methodological transparency only. All content is provisional pending Phase A execution. The locked CSI₀ specification at Phase A2 will supersede the illustrative categorisations in Extended Data 3.
