Redefining the geography of pharmaceutical innovation: Kelly Chibale as a harbinger for integrated drug discovery in Africa

Abstract

Drug discovery has historically been concentrated in high-income countries, creating a geographically asymmetric pharmaceutical ecosystem in which regions such as sub-Saharan Africa, despite bearing the highest infectious disease burden, have contributed mainly epidemiological data and clinical trial capacity rather than early-stage innovation. This imbalance is now gradually shifting, driven in large part by the leadership of Professor Kelly Chibale, who established Africa’s first fully integrated drug discovery platform, the Holistic Drug Discovery and Development (H3D) Centre at the University of Cape Town in South Africa, enabling end-to-end discovery-to-clinic progression. His team’s work spans drug discovery underpinned by medicinal chemistry innovation, institutional development, ecosystem building, and multinational collaboration, most notably through the Grand Challenges African Drug Discovery Accelerator (GC ADDA) and the Research Infrastructure Alliance to Foster Infectious Disease Research, Knowledge Sharing and Innovation (RAFIKI) consortia. Beyond advancing anti-infective drug candidates, he has led the creation of a sustainable model that integrates biology, chemistry, and pharmacology, including drug metabolism and pharmacokinetics (DMPK), and translational strategy across Africa. Through GC ADDA, this model has expanded into a distributed network linking African institutions, industry, and funders. His legacy lies in redefining the geography of drug discovery and promoting scientific sovereignty through collaboration, offering a blueprint for resilient, African-led pharmaceutical innovation in the Global South.
