Industrial and Biotechnological Applications of Mannan-Degrading Enzyme Systems

Abstract

In softwood hemicellulose and plant storage seeds such as guar gum, mannans include complex polysaccharides such as galactoglucomannan and galactomannan polysaccharides. Complete hydrolysis of these polysaccharides requires a complex, synergistic enzyme system. The core myrolysis of mannans is catalyzed by an endo-acting glycosaccharidase, β-beta-mannanase, which cleaves the mannan backbone and is supported by accessory debranching and side chain degrading enzymes, β-beta-mannosidase, β-alpha-galactosidase, and acetyl mannan esterase. Highly specialized fungi, such as Aspergillus and Trichoderma, and bacteria produce these β-beta-mannanase. These enzymes are thermally and pH stable, which is a desirable characteristic in many industrial processes. These enzyme systems are highly effective in processing lignocellulosic industrial waste, improving the digestibility of animal feeds, clarifying food and juices, and biobleaching pulp and paper. The growing focus of the field is the discovery of novel extremophiles and the engineering of stable, rapid-acting variants to facilitate more efficient enzyme systems. This review will discuss the industrial and biotechnological applications of mannan-degrading enzyme systems.Recent advances in microbial biotechnology have led to the discovery of novel mannanolytic enzymes and synergistic mechanisms, improving the efficiency of lignocellulosic biomass conversion. Fungal species such as Aspergillus and Trichoderma are especially significant producers due to their high enzymatic activity and ease of cultivation. Laboratory techniques for enzyme detection and activity quantification include colorimetric assays using locust bean gum substrates and p-nitrophenyl derivatives. Future directions in β-mannanase research focus on isolating extremophilic strains, engineering enzyme variants with improved thermal and pH stability, and optimizing recombinant production systems. Overall, mannan-degrading enzymes play a vital role in sustainable biotechnological processes, transforming renewable biomass into high-value bio-based products.
