Histological and Biochemical Comparative Study Between the Effect of Quercetin and Topiramate on Induced Diabetic Peripheral Neuropathy in Adult Male Albino Rats

Abstract

Introduction: Diabetic peripheral neuropathy (DPN) is a prevalent consequence for diabetes mellitus, significantly attributing to risk of amputation and mortality, if untreated. Aim of the Work: To emphasize the possible regenerative capacities of Quercetin and Topiramate against induced DPN in rat models.Material and Methods: 50 adult male albino rats were grouped as the follow; control group (Ia and Ib; n=10) and experimental group (II; n=40) injected once by alloxan (150mg /kg/i.p) then subdivided equally after 4 weeks from diabetic induction into 4 subgroups. Subgroup IIa remained without further treatment. Subgroup IIb received Quercetin (100mg/Kg), subgroup IIc treated with Topiramate (30mg/kg), and subgroup IId received Quercetin with Topiramate in the same doses of IIb & IIc respectively. All were given by gastric gavage for another 4 weeks then the blood samples were withdrawn for biochemical assessments. Sciatic nerves were harvested for histological analyses using H&E., Toluidine-blue, immunohistochemical staining for caspase-3 and S-100 immunoreactivity respectively. In addition to ultra-thin sections for electron microscopic examination followed by both morphometric and statistical studies.Results: Subgroup IIa emphasized altered biochemical and immunohistochemical features; significant decline in nerve fibres count with disturbed organization, abnormal myelin configurations, axonopathy and significant increase and decrease in caspase-3 and S-100 immunoreactivity. Subgroup IIb and subgroup IIc revealed comparable results to each other inform of partial improvements. Optimal improvements were clearly emphasized with combined therapy in subgroup IId.Conclusion: Concomitant administration of Quercetin and Topiramate proved to exert better regenerative results than each treatment alone in experimentally induced diabetic neuropathy
